What is bacitracin?

Bacitracin is a white to off-white fluffy powder. It is one of our China GMP-Approved APIs along with Bacitracin zinc, polymyxin b sulfate. It is soluble in water, ethanol, and pyridine. Its acidic or neutral aqueous solution is relatively stable. When pH maintains at 5-7, the bacitracin solution is stable for 4 weeks. When solution pH is greater than 9, bacitracin could degrade rapidly at room temperature.

Bacitracin, first isolated in 1945 and later commercially produced, is a mixture of related polypeptides widely used as an antibiotic in injections or topical formulations.

It is a mixture of related compounds with different antibacterial activities. Components include bacitracin A, A1, B, B1, B2, C, D, E, F, G, and X. Bacitracin A was found to have the most antibacterial activity. Bacitracin B1 and B2 have similar potencies and are approximately 90% as active as bacitracin A. Other bacitracin components including F and X do not appear to be extensively studied.

Bacitracin is manufactured by growing the bacteria Bacillus subtilis var Tracy in a container of liquid growth medium in our FDA-audited facility. After fermentation, the bacteria synthesize the antibiotic and secrete the antibiotic into the medium. Then bacitracin is extracted from the medium through chemical processing.

Bacitracin is an effective narrow-spectrum polypeptide with an antibacterial spectrum similar to penicillin and effective against Gram-positive bacteria, some Gram-negative bacteria, spirochetes, and actinomycetes. These polypeptides inhibit Gram-positive bacteria by interfering with the cell wall and peptidoglycan synthesis.

Bacitracin related ingredients​

As per Bacitracin Monographs in USP38-NF33 S2, bacitracin has 8 related ingredients: Bacitrain A, B1, B2, B3, C1, C2, C3, F, those are the impurities listed in USP. But according to wikipedia, bacitracin is made up by 11 components:A, A1, B, B1, B2, C, D, E, F, G and X.

Among the 8 substances, 4 of them are active ingredients: Bacitracin A, Bacitracin B1, Bacitracin B2, Bacitracin B3. The rest of the ingredients are regarded as impurities such as Bacitracin C1, C2, C3, F.

How is bacitracin produced?

Bacitracin is commercially manufactured by growing the bacteria Bacillus subtilis var Tracy I in a container of liquid growth medium. After a certain period, the bacteria synthesize the antibiotic and secrete the antibiotic into the medium. Then the antibiotic is extracted from the medium through chemical processes.

1. Fermentation: use Bacillus subtilis and B. licheniformis to get bacitracin solution. (Bacitracin solution is an extraction and separation solution of a Bacillus subtilis and a B-type Bacillus licheniformis fermentation broth with a potency of no less than 5000 u/ml and a conductance of less than 2000 μs/cm).

2. Adjusting PH value: an alkaline solution is added to the bacitracin solution to adjust the pH as 8 to 11, adjust pH as 5 to 7 with an inorganic acid, stir for 1 to 2 hours, and let stand for 5 to 10 hours.

3. Extraction: bacitracin is then obtained by membrane separation, drying, pulverization, and sieving.
The spray-dried bacitracin powder obtained by processing above can be used as a raw material for medicine preparations such as bacitracin injections and bacitracin polymyxin b ointments.

The total content of bacitracin A is no less than 40.0%, the total amount of bacitracin A, B1. B2 and B3 should be greater than 70.0% and the potency is no less than 60IU/MG.

What can bacitracin be used for?

Bacitracin is used for human medicine as a polypeptide antibiotic against infections. Please note, bacitracin should be used only to treat or prevent infections that are proven or strongly suspected to be caused by susceptible bacteria.

Bacitracin is primarily made into ointments as it is can cause kidney damage when used internally.

Bacitracin has been indicated in topical formulations for acute and chronic localized skin infections. It is also used intramuscularly for infantile streptococcal pneumonia and empyema. Bacitracin is also formulated as an ointment with neomycin and polymyxin B as OTC.

Bacitracin ointment formulated with neomycin and polymyxin B along with hydrocortisone is indicated for the treatment of corticosteroid-responsive dermatoses with secondary infection.

Bacitracin can also be used in pure form for those with allergies to the polymyxin B and neomycin components of the combination product.

Bacitracin is also commonly used as an aftercare antibiotic on tattoos and circumcision. It is preferred over combination products such as Neosporin because of its fewer ingredients, which lowers the chances of an allergic reaction.

Although bacitracin’s allergic cross-reaction with sulfa drugs has been occasionally reported, bacitracin-containing topical preparations is a great alternative to silver sulfadiazine (Silvadene) for burn patients with a sulfa allergy.

Bacitracin has been used to distinguish Streptococcus pyogenes from other “strep” bacteria, with S. pyogenes being sensitive to bacitracin and others resistant. In this case, bacitracin is used to distinguish S. pyogenes from other β-hemolytic streptococci.

Mupirocin is an antibacterial pseudomonic acid antibiotic produced by Pseudomonas fluorescens. It is mainly used for skin infections such as impetigo, other primary or secondary superficial skin infections, such as ecthyma, eczema, folliculitis, furunculosis, atopic dermatitis, epidermolysis bullosa, and minor wounds, burns or ulcers.

While bacitracin is used for infection-preventions, especially wounds on the skin such as cuts or burns. It is also used for infantile streptococcal pneumonia and empyema when used intramuscularly.

#2. Different working mechanism

Mupirocin works by preventing bacteria from growing. It inhibits protein synthesis in bacteria by binding to isoleucyl transfer RNA synthetase. While bacitracin is a powerful bacteria killer so it will prevent the wounds from infections.

#3 Different Dosage forms

There are mainly 4 dosage forms of Mupirocin: 1. Mupirocin Calcium Ointment 2. Mupirocin Cream 3. Mupirocin Nasal Ointment 4. Mupirocin Ointment. Dosage forms for bacitracin include bacitracin for injection, bacitracin ointment and bacitracin ophthalmic ointment.

As we can there are only topical dosage forms for mupirocin no mupirocin injections. Neither does Mupirocin have eye ointments while bacitracin has injections as well as ophthamic ointments.

The reason that mupirocin can’t be made into eye ointments is because: once mupirocin is applied to eyes, there will be severe symptoms such as burning, tearing. It will take days or even weeks for this uncomfortableness to disappear. (If contact with the eyes occurs, thoroughly wash eyes with water or go to a doctor immediately).

However, there is mupirocin ointment for intranasal use, while there are no bacitracin dosage forms suitable for intranasal uses.
